preparation | to L-ornithine hydrochloride (5.0g,29.7mmol) in aqueous sodium hydroxide solution (50ml, 0.5M) to this solution was added a solution of copper sulfate (17.8g, mmol) in water (ml), and the mixture was stirred at room temperature for 4 hours. The mixture was then cooled to 0 °c before sodium bicarbonate (5.0g,59.4mmol) and benzyl chloroformate (5.7mL,40.1mmol) were added. The pH was then adjusted to pH 9 by careful addition of aqueous sodium hydroxide (1.0M) and the mixture was stirred at room temperature for 4 hours. The resulting solid was collected by filtration, washed with water, then ether, dissolved in aqueous EDTA (0.5 ml, M) and stirred vigorously at room temperature for 14 hours. The solid was again collected by filtration and washed with water to give crude solid N'-Cbz-L-ornithine, which was used without further purification. |
